    Simultaneous implementation of magnetic resonance imaging methods for Arterial Spin Labeling (ASL) and Blood Oxygenation Level Dependent (BOLD) imaging makes it possible to quantitatively measure the changes in cerebral blood flow (CBF) and cerebral oxygen metabolism (CMRO(2)) that occur in response to neural stimuli. To date, however, the range of neural stimuli amenable to quantitative analysis is limited to those that may be presented in a simple block or event related design such that measurements may be repeated and averaged to improve precision. Here we examined the feasibility of using the relationship between cerebral blood flow and the BOLD signal to improve dynamic estimates of blood flow fluctuations as well as to estimate metabolic-hemodynamic coupling under conditions where a stimulus pattern is unknown. We found that by combining the information contained in simultaneously acquired BOLD and ASL signals through a method we term BOLD Constrained Perfusion (BCP) estimation, we could significantly improve the precision of our estimates of the hemodynamic response to a visual stimulus and, under the conditions of a calibrated BOLD experiment, accurately determine the ratio of the oxygen metabolic response to the hemodynamic response. Importantly we were able to accomplish this without utilizing a priori knowledge of the temporal nature of the neural stimulus, suggesting that BOLD Constrained Perfusion estimation may make it feasible to quantitatively study the cerebral metabolic and hemodynamic responses to more natural stimuli that cannot be easily repeated or averaged

    Absorption, Excretion, and Biotransformation of Dimethyl Sulfoxide in Man and Miniature Pigs After Topical Application as an 80% Gel

    The absorption, excretion, and biotransformation of dimethyl sulfoxide (DMSO) 80% gel, DEMASORB®, was studied in man and in miniature pigs. DMSO 80% gel (15 cc, t.i.d.) was applied topically to the elbows of human subjects and allowed to remain there for 30 minutes after each application. Under these conditions, daily absorption of DMSO 80% gel ranged from 25 to 40% of the total dose. DMSO 80% gel (15 g, t.i.d.) was completely absorbed within 4 hours after application to the shaved backs of miniature pigs. Both man and miniature pig transformed DMSO to dimethylsulfone (DMSO2) and dimethylsulfide (DMS). DMSO and DMSO2 were excreted in the urine, whereas DMS was eliminated in the expired air. In man, the relative amounts of DMSO and DMSO2 in the plasma were similar to those found in the urine. The biological half-life of DMSO2 in both the plasma and urine of man was 2.5 to 3 days. Urinary excretion of DMSO plus DMSO2 ranged from 9 to 35% of the dose in both man and miniature pigs; only 1.6% of the dose was present in the feces of miniature pigs. Whereas DMSO2 was the main excretory product in the urine of man, DMSO was the major component in the urine of miniature pigs

    Abundance and stomach content analysis of threadfin shad in Lake Mead, Nevada: Do invasive quagga mussels affect this prey species?

    Lake Mead, Nevada is the largest reservoir by volume in the United States, as well as a popular sport fishing destination. In January 2007, the invasive quagga mussel Dreissena rostriformis bugensis (Andrusov, 1897) was discovered in the reservoir and concerns began to arise about potential alterations to the aquatic environment. The Lake Mead sport fishery is reliant on the sustainability of prey species like the threadfin shad [Dorosoma petenense (Günther, 1867)]. This study examined 20-year trends in historic abundance of the threadfin shad, before, and shortly after, the discovery of quagga mussels in the system. Larval shad trawl data collected in Las Vegas Bay and Overton Arm portions of Lake Mead were analyzed in the present study. Two-way analysis of variances showed that the abundance of this prey fish has not changed following quagga mussel invasion (P \u3e 0.05). Post-quagga mussel discovery collections of adult threadfin shad from Las Vegas Bay ranged from 113 to 212 mm total length (TL) (mean = 184 mm; n = 170). Shad from Overton Arm ranged from 131 to 197 mm TL (mean = 150 mm; n = 27). Stomach contents were analyzed. The proportion of cladocerans in stomach contents differed significantly from spring to winter in Las Vegas Bay (P = 0.008); whereas the proportion of copepods did not show statistically relevant differences regarding seasonality (P = 0.562). Initial trends in lower trophic level dynamics in response to quagga mussel invasion has yet to reveal significant effects in Las Vegas Bay or Overton Arm. Long-term monitoring on threadfin shad is needed to evaluate potential impacts from invasive quagga mussels in Lake Mead

    Pharmacy Students’ Performance and Perceptions in a Flipped Teaching Pilot on Cardiac Arrhythmias

    Objective. To implement the flipped teaching method in a 3-class pilot on cardiac arrhythmias and to assess the impact of the intervention on academic performance and student perceptions. Design. An intervention group of 101 first-year pharmacy students, who took the class with the flipped teaching method, were supplied with prerecorded lectures prior to their 3 classes (1 class in each of the following subjects: basic sciences, pharmacology, and therapeutics) on cardiac arrhythmias. Class time was focused on active-learning and case-based exercises. Students then took a final examination that included questions on cardiac arrhythmias. The examination scores of the intervention group were compared to scores of the Spring 2011 control group of 105 first-year students who took the class with traditional teaching methods. An online survey was conducted to assess student feedback from the intervention group. Assessment. The mean examination scores of the intervention group were significantly higher than the mean examination scores of the control group for the cardiac arrhythmia classes in pharmacology (with 89.6 6 2.0% vs 56.8 6 2.2%, respectively) and therapeutics (89.2 6 1.4% vs 73.7 6 2.1%, respectively). The survey indicated higher student satisfaction for flipped classes with highly rated learning objectives, recordings, and in-class activities. Conclusion. Use of the flipped teaching method in a 3-class pilot on cardiac arrhythmias improved examination scores for 2 of the 3 classes (pharmacology and therapeutics). Student satisfaction was influenced by the quality of the learning objectives, prerecorded lectures, and inclass active-learning activities

    An operations and command systems for the extreme ultraviolet explorer

    About 40% of the budget of a scientific spacecraft mission is usually consumed by Mission Operations & Data Analysis (MO&DA) with MO driving these costs. In the current practice, MO is separated from spacecraft design and comes in focus relatively late in the mission life cycle. As a result, spacecraft may be designed that are very difficult to operate. NASA centers have extensive MO expertise but often lessons learned in one mission are not exploited for other parallel or future missions. A significant reduction of MO costs is essential to ensure a continuing and growing access to space for the scientific community. We are addressing some of these issues with a highly automated payload operations and command system for an existing mission, the Extreme Ultraviolet Explorer (EUVE). EUVE is currently operated jointly by the Goddard Space Flight Center (GSFC), responsible for spacecraft operations, and the Center for Extreme Ultraviolet Astrophysics (CEA) of the University of California, Berkeley, which controls the telescopes and scientific instruments aboard the satellite. The new automated system is being developed by a team including personnel from the NASA Ames Research Center (ARC), the Jet Propulsion Laboratory (JPL) and the Center for EUV Astrophysics (CEA). An important goal of the project is to provide AI-based technology that can be easily operated by nonspecialists in AI. Another important goal is the reusability of the techniques for other missions. Models of the EUVE spacecraft need to be built both for planning/scheduling and for monitoring. In both cases, our modeling tools allow the assembly of a spacecraft model from separate sub-models of the various spacecraft subsystems. These sub-models are reusable; therefore, building mission operations systems for another small satellite mission will require choosing pre-existing modules, reparametrizing them with respect to the actual satellite telemetry information, and reassembling them in a new model. We briefly describe the EUVE mission and indicate why it is particularly suitable for the task. Then we briefly outline our current work in mission planning/scheduling and spacecraft and instrument health monitoring

    Trace-element and physical response to melt percolation in Summit (Greenland) snow

    Surface melt on a glacier can perturb the glaciochemical record beyond the natural variability. While the centre of the Greenland ice sheet is usually devoid of surface melt, many high- Arctic and alpine ice cores document frequent summertime melt events. Current hypotheses interpreting melt-affected ice-core chemistry rely on preferential elution of certain major ions. However, the precise nature of chemistry alteration is unknown because it is difficult to distinguish natural variability from melt effects in a perennially melt-affected site. We use eight trace-element snow chemistry records recovered from Summit, Greenland, to study spatial variability and melt effects on insoluble trace chemistry and physical stratigraphy due to artificially introduced meltwater. Differences between non-melt and melt-affected chemistry were significantly greater than the spatial variability in chemistry represented by nearest-neighbour pairs. Melt-perturbed trace elements, particularly rare earth elements, retained their seasonal stratigraphies, suggesting that trace elements may serve as robust chemical indicators for annual layers even in melt-affected study areas. Results suggest trace-element transport via meltwater percolation will deposit eluted material down-pit in refrozen areas below the nearest-surface chemistry peak. In our experiments, snow chemistry analyses are more sensitive to melt perturbations than density changes or unprocessed near-infrared digital imagery

    Accumulating evidence suggests that self-renewal and differentiation capabilities reside only in a subpopulation of tumor cells, termed cancer stem cells (CSCs), whereas the remaining tumor cell population lacks the ability to initiate tumor development or support continued tumor growth. In head and neck squamous cell carcinoma (HNSCC), as with other malignancies, cancer stem cells have been increasingly shown to have an integral role in tumor initiation, disease progression, metastasis and treatment resistance. In this paper we summarize the current knowledge of the role of CSCs in HNSCC and discuss the therapeutic implications and future directions of this field

    High efficiency, low distortion 3D diffusion tensor imaging with variable density spiral fast spin echoes (3D DW VDS RARE)

    We present an acquisition and reconstruction method designed to acquire high resolution 3D fast spin echo diffusion tensor images while mitigating the major sources of artifacts in DTI-field distortions, eddy currents and motion. The resulting images, being 3D, are of high SNR, and being fast spin echoes, exhibit greatly reduced field distortions. This sequence utilizes variable density spiral acquisition gradients, which allow for the implementation of a self-navigation scheme by which both eddy current and motion artifacts are removed. The result is that high resolution 3D DTI images are produced without the need for eddy current compensating gradients or B_0 field correction. In addition, a novel method for fast and accurate reconstruction of the non-Cartesian data is employed. Results are demonstrated in the brains of normal human volunteers

    Racial and Ethnic Variation in Lipoprotein (a) Levels among Asian Indian and Chinese Patients

    Background. Lipoprotein (a) [Lp(a)] is an independent risk factor for cardiovascular disease (CVD) in Non-Hispanic Whites (NHW). There are known racial/ethnic differences in Lp(a) levels, and the association of Lp(a) with CVD outcomes has not been examined in Asian Americans in the USA. Objective. We hypothesized that Lp(a) levels would differ in Asian Indians and Chinese Americans when compared to NHW and that the relationship between Lp(a) and CVD outcomes would be different in these Asian racial/ethnic subgroups when compared to NHW. Methods. We studied the outpatient electronic health records of 2022 NHW, 295 Asian Indians, and 151 Chinese adults age ≥18 y in Northern California in whom Lp(a) levels were assessed during routine clinical care from 2001 to 2008, excluding those who had received prescriptions for niacin (14.6%). Nonparametric methods were used to compare median Lp(a) levels. Significance was assessed at the P < .0001 level to account for multiple comparisons. CVD outcomes were defined as ischemic heart disease (IHD) (265 events), stroke (122), or peripheral vascular disease (PVD) (87). We used logistic regression to determine the relationship between Lp(a) and CVD outcomes. Results. Both Asian Indians (36 nmol/L) and NHW (29 nmol/L) had higher median Lp(a) levels than Chinese (22 nmol/L, P ≤ .0001 and P = .0032). When stratified by sex, the differences in median Lp(a) between these groups persisted in the 1761 men (AI v CH: P = .001, NHW v CH: P = .0018) but were not statistically significant in the 1130 women (AI v CH: P = .0402, NHW v CH: P = .0761). Asian Indians (OR = 2.0) and Chinese (OR = 4.8) exhibited a trend towards greater risk of IHD with high Lp(a) levels than NHW (OR = 1.4), but no relationship was statistically significant. Conclusion. Asian Indian and NHW men have higher Lp(a) values than Chinese men, with a trend toward, similar associations in women. High Lp(a) may be more strongly associated with IHD in Asian Indians and Chinese, although we did not have a sufficient number of outcomes to confirm this. Further studies should strive to elucidate the relationship between Lp(a) levels, CVD, and race/ethnicity among Asian subgroups in the USA
